Methamphetamine (MA), a psychostimulant, has become a serious problem in recent years. It is one of the most common psychostimulants in the world. In the Czech Republic, ecstasy is the most common non-cannabis drug, followed by hallucinogenic mushrooms, LSD, MA, cocaine and finally heroin. The prevalence of the use of all addictive substances is highest in the age group of 15–34 years. Approximately 17.2% of registered drug addicts, both men and women, in the Czech Republic use MA as their drug of first choice. This group consists mostly of unemployed and MA-dependent women (85%). Almost half of addicted women switched to MA from other drugs during pregnancy. Psychostimulants, such as amphetamine and its synthetic derivative MA, induce feelings of calmness and happiness, suppressing anxiety and depression. When MAs are abused for long periods, it mimics the symptoms of mania and can lead to the development of psychosis. MA is often abused because of its anorectic effects, ease of preparation, and low cost compared to heroin and cocaine. There is considerable variation in user sensitivity to the stimulant, with reactions to MA varying from person to person. Molecular mechanisms associated with variable response among users may explain bipolar disorder and addiction-related psychosis. Currently, there is limited information regarding the genetic mechanisms associated with these diseases and the transmission of drug addiction. Thus, animal models of drug addiction are significant sources of information and resources for research on these issues. The purpose of this review is to summarize the mechanism of action of methamphetamine and its effects on pregnant addicted women and their children, including a detailed description of the anatomical structures involved.

Addiction is a set of behavioral, cognitive, and physiological states. The main reason for its development is repeated use of addictive substances; typical symptoms include a strong preference for repeated use of the substance, altered consciousness after use, and continued use despite harmful consequences. In addition, drug use takes priority over other activities and commitments, tolerance builds gradually, and when substance use is stopped, somatic withdrawal sometimes occurs. A common motive for substance abuse is curiosity, which can eventually lead to addiction. Many consumers first try a drug to find out “what it is” [1]. Other possible motives include stress or problems that the person is unable to cope with in any other way, leading to drug use as a means of escaping reality. Drug addiction is a global problem. It directly affects a large part of society, and its indirect effect on the families and surroundings of users is even more significant (Fig. 1).

An important factor in reducing drug use is prevention. In the prevention of addiction and use of alcohol and non-alcoholic drugs, the World Health Organization (WHO) identifies three goals: (1) primary prevention, that is, the prevention of drug use by those who have not had direct contact with drugs; (2) secondary prevention, i.e. prevention of addiction in drug users; (3) tertiary prevention, i.e. the prevention of serious and long-term health and social problems caused by drug use, including both resocialization and social rehabilitation and measures to reduce the transmission of infectious diseases during intravenous drug use (e.g. programs harm reduction) [5].

The initial phase of drug addiction, that is, when the user experiences euphoria and still detects the effects of the drug, is called the phase of experimental use. During the social use phase, the user begins to take the drug more regularly, often at higher doses; during this phase, addiction begins to manifest itself, and the drug gradually becomes the source of various problems. With daily use, the drug begins to affect things like daily routines, moral values, work, and friendships. Ultimately, the user loses control over his addiction [6]. Chemical dependence (ie, needing more of the substance to achieve the same effect) is the final stage of addiction, often ending in fatal overdose or complete organ failure.
