Introduction • Hemoglobin (Hb) is a metalloprotein found in red blood cells. • Red blood cells carry oxygen throughout the body. • All vertebrates except fish have hemoglobin in their red blood cells as an oxygen carrier. • Hemoglobin makes up 96% of the dry weight of red blood cells and contains iron. • The normal hemoglobin level for a normal male adult is 13.8 – 17.0 g/dL. An adult woman’s (non-pregnant) hemoglobin level should be between 12.0 and 15.0 g/dL. Hemoglobin Function • Hemoglobin is an oxygen carrier. • Hemoglobin is a carbon dioxide carrier. • Hemoglobin maintains the shape and color of red blood cells. • Hemoglobin acts as a buffer. • Hemoglobin interacts with other ligands. • Hemoglobin breakdown leads to accumulation of physiologically active catabolites. Oxygen Transport • The main function of hemoglobin is to transport oxygen from the lungs to all tissues of the body. • The oxygen binding capacity of hemoglobin is 1.34 mL O2 per gram. • The affinity of hemoglobin for oxygen is achieved by Fe2+ ions. Each Fe2+ can bind to one oxygen molecule. • Oxygen bond oxidizes Fe2+ to Fe3+. • Oxygen-binding hemoglobin is called oxyhemoglobin. When blood reaches oxygen-deficient tissues, oxygen separates from the hemoglobin and diffuses into the tissues. • When O2 is removed, iron changes to its reduced form.
