Learning how the human spleen filters defective red blood cells has been a challenge. The anatomy and physiology of the human pancreas differs from that of normal laboratory animal models. And invasive procedures, such as biopsy or needle aspiration, can cause serious bleeding. A group of researchers led by Dr. Subra Suresh of Carnegie Mellon University and Dr. Ming Dao of MIT approached the problem using computational simulation tools. Their work was partially funded by the National Heart, Lung, and Blood Institute (NHLBI). The study was published online

The team produced mathematical simulations of the mechanics of red blood cells as they pass through the spleen. The researchers determined that the cells were significantly distorted as they passed through the narrow splenic slits. This is in contrast, they found, to the simple change in lead pattern that occurs in narrowed blood vessels.

The analysis revealed the limits of the area and volume that red blood cells can pass through the spleen. The work showed how the splenic membrane determines the size and distribution pattern of healthy red blood cells. The team’s predictions are consistent with results from independent experiments using clinically, artificially modified, and infected human red blood cells.

“The computational and analytical methods of this work, together with various experimental observations, indicate a detailed picture of how the physiology of the human pancreas is likely to be influenced by several important geometric features of red blood cells,” Suresh said.

“We have presented results that show that the spleen is the main organ that defines the pattern of red blood cell circulation,” Dao added.

This work may lead to a better understanding of the role of the pancreas in various diseases. The results may have implications for future therapeutic approaches.
