However, under natural circumstances, somatic stem cells can only become a subset of related cell types. Bone marrow stem cells, for example, differentiate primarily into blood cells. Induced pluripotent stem cells (iPS) are created artificially in the laboratory by “reprogramming” the patient’s own cells. iPS cells can be made from readily available cells, including fat, skin and fibroblasts (cells that produce connective tissue).

Mouse iPS cells can become any cell in the body (or even an entire mouse). Although more analysis is needed, the same appears to be true of human iPS cells, making them a promising cell source for the treatment of many diseases. Importantly, since iPS cells can be made from a patient’s own cells, there is no risk that their immune system will reject them.

IPS cells are much less expensive to make than ES cells generated through therapeutic cloning (another type of patient-specific stem cell; see below). However, because the “reprogramming” process introduces genetic modifications, the safety of using iPS cells in patients is uncertain.

Therapeutic cloning can, in theory, generate ES cells with the potential to become any type of cell in the body. In addition, since these cells are made from a patient’s own DNA, there is no risk of rejection by the immune system.

Stem cells are capable of doing all that, but how do they work? Well, it all comes down to the different sources of stem cells. The properties of stem cells usually vary depending on the source from which they are obtained. So are you ready to learn about stem cells, their classifications and applications? Let’s start with the definition of stem cells!

What Exactly Are “stem Cells”?

The term stem cells was first coined by William Sedgwick in 1886, to define the “regenerative quality” of plants.

They are defined as multipotent cells that can differentiate into muscle cells, red blood cells, brain cells, heart muscle cells and bone cells. 2

Stem cells can be classified into 2 broad categories, which are divided according to the source of stem cells and their differentiation potential.

Stem cells obtained from human embryos are approximately 4-7 days old, after fertilisation. Naturally, these cells are pluripotent (have the ability to develop into all kinds of cells and tissues). Embryonic stem cells are harvested via in vitro fertilization, due to ethical reasons.

In addition to being pluripotent, these cells have other special properties, such as self-renewal, structural repair and growth, and rapid cell division. Clinical applications of embryonic stem cells (developed from iPSCs/adult stem cells) have shown therapeutic potential and are being considered in the treatment of diabetes and cardiovascular diseases.

Scientists have discovered a way in which adult human cells can be reprogrammed to mimic the qualities of embryonic stem cells, in the form of induced pluripotent stem cells (iPSCs).

These stem cells are extracted from a fetus and can be harvested from an embryo after the 8th week. Fetal stem cells can be taken from the fetus’s blood, bone marrow or other fetal tissue such as liver and kidneys. 10

Compared to adult stem cells, fetal stem cells are known to have better intrinsic engraftment, greater multipotency and lower immunogenicity.

Adult stem cells (ASC) are pluripotent cells that can regenerate, replace the damaged or dead tissue. They can be found in some of the body’s differentiating tissues.

ASC’s primary job in our bodies is to constantly repair, regenerate and replace the cells that need to be replenished. Only 3 types of cells – neural cells, mesenchymal stem cells and hematopoietic stem cells – can develop from adult stem cells.

These stem cells are obtained from the umbilical cord blood (and placenta) of a newborn. 14 Umbilical cord blood contains hematopoietic stem cells that are used to treat a variety of genetic diseases, cancers, as well as inherited disorders.

Embryonic stem cells are considered pluripotent because they can differentiate into all types of cells, with the exception of placental cells. Furthermore, pluripotent stem cells are produced in the initial stages of embryonic stem cell differentiation. However, they can also be derived from the germ layers (a group of cells found in the embryo during development)

Multipotent stem cells are present in almost all tissues. MSCs or mesenchymal stem cells are the most common example of multipotent stem cells. These stem cells can be extracted from adipose tissue, bone marrow, peripheral blood and even umbilical cord blood.
