Chlorophyll is a green molecule found in photosynthetic organisms, which plays a key role in absorbing light and converting it into chemical energy, the first step of photosynthesis. In recent years, several research groups have reported the rapid response of chlorophyll in microdroplets. Solvent evaporation or low voltage were cited as the main causes of acceleration. However, this study conducted experiments challenging existing assumptions and found that the limiting effect of the ical space of micro-sized droplets is the reason for the rapid response.

The team focused on the chemical reactions of chlorophyll to reveal the secrets that control the absorption and transfer of solar energy. Under acidic conditions, the chlorophyll demethylation reaction occurs when magnesium ions in the center of chlorophyll are replaced by hydrogen ions. The importance of this reaction in photosynthesis has been overlooked because the rate of the demetalation reaction in bulk solution was much slower than the rate of absorption and transfer of solar energy in many experiments.

In living organisms, a variety of biochemical reactions take place in completely confined spaces. Photosynthetic organelles, chloroplasts of plants and small structures called ‘granules’ in chloroplasts absorb light. The team observed biochemical reactions by creating microdroplets to see how chlorophyll reacts in a similar environment inside plants.

The research team collided water droplets containing chlorophyll at high speed with water droplets containing hydrochloric acid to form micro-sized fused droplets. They then recorded the kinetics of acid-induced chlorophyll demethylation by controlling the travel distance of the fused microdroplets.

As a result, the researchers found that chlorophyll demetalation occurs within several tens of microseconds, about 1000 times faster than the process in bulk solution. This result is thought to be due to the limitation of the ical space of the microdroplets as well as the surface effect of the droplets themselves.

“When chlorophyll is oxidized, it loses its photosynthesis function. However, the demetallation reaction can protect chlorophyll because it prevents the oxidation of chlorophyll,” said Hong-Gil Nam.

He added, “This study suggests that the demetalation reaction of chlorophyll may be a novel mechanism to protect or control photosynthetic efficiency of photosynthetic organisms, and that this reaction, unlike traditional methods, requires no enzymatic activity. The process can be quite fast.”

The team expects the discovery to contribute to further studies of the elements and methods of more efficient photosynthesis.
